Key Points

  • CCR3+ cell numbers and marker expressions are significantly altered in allergic asthma and allergic rhinitis patients
  • CD203c emerged as a more selective basophil activation marker compared to CD63
  • Combined CCR3 and CD203c markers may offer a reliable diagnostic approach for allergic respiratory conditions

Summary

This study investigated basophil and eosinophil marker expressions in peripheral blood of patients with allergic asthma (AA) and combined allergic rhinitis and asthma (ARA). Using flow cytometry, researchers comprehensively analyzed CCR3, CD63, CD203c, and FcεRIα markers in granulocyte and peripheral blood mononuclear cell (PBMC) populations, revealing significant alterations in marker expressions associated with allergic inflammatory conditions.

The research demonstrated markedly increased CCR3+ cell numbers in granulocytes of AA and ARA patients, with up to 96% eosinophils and 12.8% basophils. Notably, the mean fluorescent intensity of activation markers CD203c, FcεRIα, and CD63 was consistently enhanced in both granulocytes and PBMC. Plasma levels of inflammatory mediators (EDN, LTC4, PGD2) correlated well with CCR3+ granulocyte numbers, suggesting these markers could provide valuable diagnostic insights into allergic respiratory diseases.
